Description

In our present research work a mixture of house-hold agricultural wastes were utilize as fermentation medium for solid state fermentation to make the process more economical. In this study, adsorption property of activated charcoal was utilized, which resulted in maximum decolourization along with retention of an appreciable activities for the produced alpha amylase(11261.5280 U/gds) & amyloglucosidase (193.3217 U/gds ) when compared to control. Purification by chilled acetone precipitation method was 56.88 fold for enzyme alpha amylase and 4.57 fold for amyloglucosidase. This was the highest fold of purification achieved for alpha amylase and amyloglucosidase of Aspergillus oryza by chilled acetone precipitation methods reported till date. This also signifies the novelty of the work. Finally, both the purified enzymes were lyophilized for increasing the shelf-life with practically little loss of activity. Preserved enzymes may found an industrial use, which again signifies the novelty of the work.
